Novel Drugs Recognized as a 'Pivotal Moment' in Addressing Drug-Resistant Gonorrhea

The first new treatments for gonorrhoea in decades are being viewed as a "major milestone" in the battle against increasingly resistant strains of the pathogen, according to scientists.

A Global Public Health Issue

Cases of gonorrhoea are increasing worldwide, with data suggesting more than 82 million new cases each year. Notably increased rates are reported in Africa and countries within the WHO's Western Pacific region, which spans from China and Mongolia to New Zealand. In England, cases have hit a record high, while figures across Europe in 2023 were three times higher compared to those in 2014.

“The authorization of novel therapies for gonorrhoea is an important and timely step in the reality of growing infection rates, increasing antimicrobial resistance and the very limited available drugs presently on offer.”

Health officials are particularly alarmed about the increase in drug-resistant strains. The WHO has listed it as a "high-priority threat". A tracking program revealed that the effectiveness of standard treatments like ceftriaxone and cefixime jumped significantly between 2022 and 2024.

Two New Treatment Options Receive Authorization

Zoliflodacin, alternatively called a brand name, was cleared by the US FDA in December for combating gonorrhoea. This STI can lead to significant complications, including the inability to conceive. Experts believe that targeted use of this new drug will help hinder the development of resistance.

Another new antibiotic, developed by the pharmaceutical company GSK, gained clearance in the same week. This medication, which is also used to treat UTIs, was proven in research to be effective against antibiotic-resistant forms of the gonorrhoea bacteria.

A Unique Development Model

Zoliflodacin stemmed from a innovative non-profit model for medication research. The charitable organization GARDP partnered with the pharmaceutical company its industry partner to see it through.

“This authorization represents a huge turning point in the therapy of highly resistant gonorrhoea, which until now has been evolving faster than medical innovation.”

Research Study Results and Worldwide Availability

According to findings detailed in a major medical journal, zoliflodacin cured over nine in ten of cases of the STI. This places it at an equal footing with the current standard treatment, which uses an injection and a pill. The trial enrolled hundreds of volunteers from various regions including Belgium, the Netherlands, South Africa, Thailand and the US.

As part of the agreement of its collaboration, GARDP has the authority to license and sell the drug in numerous regions with limited resources.

Doctors treating patients have expressed optimism. Having a single-dose, oral treatment like this is hailed as a "critical tool" for gonorrhoea control. This is deemed vital to lessen the impact of the infection for individuals and to stop the proliferation of highly drug-resistant gonorrhoea worldwide.
